Dephosphorylated Synapsin I Anchors Synaptic Vesicles to Actin Cytoskeleton: An Analysis by Videomicroscopy

چکیده

Synapsin I is a synaptic vesicle-associated protein which inhibits neurotransmitter release, an effect which is abolished upon its phosphorylation by Ca2÷/calmodulin-dependent protein kinase II (CaM kinase ]I). Based on indirect evidence, it was suggested that this effect on neurotransmitter release may be achieved by the reversible anchoring of synaptic vesicles to the actin cytoskeleton of the nerve terminal. Using video-enhanced microscopy, we have now obtained experimental evidence in support of this model: the presence of dephosphorylated synapsin I is necessary for synaptic vesicles to bind actin; synapsin I is able to promote actin polymerization and bundling of actin filaments in the presence of synaptic vesicles; the ability to cross-link synaptic vesicles and actin is specific for synapsin I and is not shared by other basic proteins; the cross-linking between synaptic vesicles and actin is specific for the membrane of synaptic vesicles and does not reflect either a non-specific binding of membranes to the highly surface active synapsin I molecule or trapping of vesicles within the thick bundles of actin filaments; the formation of the ternary complex is virtually abolished when synapsin I is phosphorylated by CaM kinase II. The data indicate that synapsin I markedly affects synaptic vesicle traffic and cytoskeleton assembly in the nerve terminal and provide a molecular basis for the ability of synapsin I to regulate the availability of synaptic vesicles for exocytosis and thereby the efficiency of neurotransmitter

Synapsin I: an actin-bundling protein under phosphorylation control

Synapsin I is a neuronal phosphoprotein comprised of two closely related polypeptides with apparent molecular weights of 78,000 and 76,000. It is found in association with the small vesicles clustered at the presynaptic junction. Its precise role is unknown, although it probably participates in vesicle clustering and/or release. Synapsin I is known to associate with vesicle membranes, microtubu...

Interaction of synapsin I with actin and SSVs: differential regulation by calmodulin.

The synapsin family of proteins has been implicated in the formation and retention of synapses. The four mammalian synapsins (I4 polypeptide chain size 74 kDa; Ib, 70 kDa; IIa, 63 kDa; and IIb, 52 kDa) are derived from two genes (I and I), the a and b isoforms being formed by differential mRNA splicing (Sudhof, 1989). These abundant proteins are peripheral to the membranes of small synaptic ves...

Synapsin and synaptic vesicle protein expression during embryonic and post-natal lens fiber cell differentiation.

PURPOSE Reorganization of cytoskeleton and membrane biogenesis are dynamically coordinated during lens fiber cell differentiation and development to produce an organ with precise dimensions and optical properties. Cargo vesicle trafficking is fundamental to cell elongation and has also been implicated in degenerative disease mechanisms. Alzheimer precursor protein (AbetaPP) acts with kinesin, s...

Characterization of synapsin I fragments produced by cysteine-specific cleavage: a study of their interactions with F-actin

Synapsin I is a neuron-specific phosphoprotein that is concentrated in the presynaptic nerve terminal in association with the cytoplasmic surface of synaptic vesicles. It has been demonstrated to bundle F-actin in a phosphorylation-dependent manner in vitro, a property consistent with its proposed role in linking synaptic vesicles to the cytoskeleton and its involvement in the regulation of neu...
